A search of the 24,344 ESTs of the International Triricae EST Cooperative database identified 16 barley and 50 wheat ESTs having sequence similarity to P450 genes. Primers have been designed to amplify 30 of these sequences targeting the 3’ untranslated regions where possible. In barley at least 4 P450 genes are being surveyed for SNPs including genes with homology to obtusifoliol 14 alpha-demethylase, ferulate 5-hydroxylase and cinnamate 4-hydroxylase. SNPs that discriminate between barley varieties have been identified. A number of P450 sequences from wheat are also being surveyed. At least one primer pair designed to amplify a P450 sequence from barley also amplifies products from wheat. The utility of P450 and other ESTs to identify SNPs between wheat varieties and between barley varieties is being examined.
