BARLEY ESTS: A RESOURCE FOR NOVEL MARKERS

With the development of EST sequencing projects, a wealth of sequence information is being generated allowing to genetically map these ESTs.  In order to initiate a systematic approach to develop an EST-map of barley (Hordeum vulgare L.), we have developed RFLP, SSR and SNP markers. RFLPs are being identified and mapped using conventional techniques while a set of SSRs was identified by mining the EST-database for corresponding motifs. SNPs were identified between the parental genotypes of our three mapping populations by comparative sequencing. Once a SNP has been established, it is mapped by denaturing high-performance liquid chromatography (DHPLC). Employing these three assays, we plan to place over 1000 genes onto a consensus EST map of barley with preliminary results being presented.
